چکیده

Sphingomyelin, ceramide, cerebroside, and cerebroside sulfate from beef brain were isolated by column chromatography in quantitative yield and in pure form. The fatty acid composition of each sphingolipid was determined by gas-liquid chromatography (GLC). Sphingomyelin contained only nonhydroxy fatty acids, while cerebroside, cerebroside sulfate, and ceramide contained both nonhydroxy and hydroxy fatty acids. The nonhydroxy fatty acid composition of each sphingolipid was similar, except that more stearic acid was present in sphingomyelin and ceramide than in cerebroside and cerebroside sulfate. The hydroxy fatty acid compositions of cerebroside and cerebroside sulfate were, with minor exceptions, similar to each other and to the corresponding nonhydroxy fatty acid compositions. In both the nonhydroxy and hydroxy fatty acid series, both odd and even chain saturated and monounsaturated fatty acids containing 14 to 26 carbons were present.

In Vivo Conversions of Cerebroside and Ceramide in Rat Brain.

Lignoceric a~id-1-C'~ was used to make lignoceroyl psychosine (kerasin) and stearic acid-l-Cl4 to make stearoyl sphingosine (ceramide) . Emulsions of the labeled sphingolipids were injected directly into the brains of young rats and the resultant lipoidal metabolic products were examined for radioactivity. Most of the metabolized C14 was found in the ester-linked fatty acids, particularly in th...

Changes in sphingosine and fatty acid components of the gangliosides in developing rat and human brain.

Rat brain increases in weight after birth in three stages: (I) rapidly for the first 2 weeks, (II) at a lower rate from 2 to 5 weeks, and (III) at a still lower rate from 5 weeks to 5 months. During the succeeding period, designated IV, it maintains constant weight up to 1 year of age. Brain ganglioside content increased linearly during I and II, more slowly during III, and diminished during IV...

Brain Lipids: I. Quantification and Fatty Acid Composition of Cerebroside Sulfate in Human Cerebral Gray and White Matter.

Cerebroside sulfate was isolated by column chromatography from infant whole brains and adult human cerebral grey and white matter. Fatty acid analyses, performed by gas-liquid chromatography (GLC), of normal and ahydroxy fatty acids obtained after hydrolysis revealed that cerebroside sulfate contains long-chain fatty acids ranging from 14-26 carbons both in the normal and hydroxy series. In mat...

Formation of free fatty acid and ceramide during brain handling: lability of sphingomyelin.

Intact brain and brain homogenates readily form free fatty acids and ceramides, even in the cold during subcellular isolation procedures. The fatty acid formation is slightly stimulated by chelators and might be due to phospholipid hydrolysis by lysosomal phospholipases. The ceramide formation is accompanied by loss of sphingomyelin and is apparently due to the action of neutral, metal ion-acti...

Effect of cyclodextrins on the solubilization of lignoceric acid, ceramide, and cerebroside, and on the enzymatic reactions involving these compounds.

alpha-Cyclodextrin at concentrations of 1-8 mM helps dissolve, in aqueous solution, fatty acids such as lignoceric, stearic, and palmitic, and complex lipids such as ceramide and cerebroside that contain these acids. Formation of an inclusion complex was indicated on examination of the solution by gel filtration. alpha-Cyclodextrin strikingly increased synthesis of ceramide from sphingosine and...
